l General Information

Product Name

Penicillinase from Bacillus cereus

General deion

Penicillinase from Bacillus cereus has been shown to contain three distinct species when purified and examined for homogeneity by sedimentation analysis, amino-terminal analysis, and vertical acrylamide gel electrophoresis.

Synonym

β-Lactamase I from Bacillus cereus, β-Lactamase II, Cephalosporinase, Penicillin amido-β-lactam hydrolase

Assay

1,500-3,000 units/mg protein (using benzylpenicillin); Protein, ~10%

CAS Number

9073-60-3

MDL number

MFCD00131819

Suitability

BioReagent

l Physical and Chemical Information

Appearance

White to Light Yellow/Tan lyophilized powder

Physical form

Lyophilized powder containing phosphate and citrate buffer salts

l Biological Information

Biochem/Physiol Actions

Penicillinase specifically catalyzes the hydrolysis of β-lactam ring of penicillin.

Application

Penicillinase can be used to reduce levels of penicillin in patients with severe penicillin reactions. Used in the production of penicillin. Penicillinase from Bacillus cereus has been used in a comparison study with penicillinase from Bacillus subtillis where the Michaelis constant, shape of the Ph/activity curve and immunological properties were shown to be significantly different.

Used in the production of penicillin.

Preparation Note

Lyophilized powder containing phosphate and citrate buffer salts; The product is soluble (10 mg/ml) in 0.1 M Tris HCl, pH 7.0, containing 0.1% BSA. The solution can be filtered through a 0.2 mm, low protein-binding membrane.

Unit Definition

One unit will hydrolyze 1.0 μmole of indicated substrate per min at pH 7.0 at 25 °C. Sold as benzylpenicillin units. This International Unit (using benzylpenicillin as substrate) is approximately equal to 600 Levy or 75 Pollock units.

l Storage

Storage temp.

2-8°C
