Biochem/Physiol Actions

Cleavage of DNA by Nuclease micrococcal is greater at the 5′ sides of A or T nucleotides than at G or C.

Hydrolyzes 5′-phosphodiester bonds of DNA.

Application

Nuclease from Staphylococcus aureus has been used in a study to assess coagulase and heat-resistant strains found in animals. It has also been used in a study to investigate the  characteristic of two genes in S. aureus that encode two thermostable nucleases.

Preparation Note

Reconstitute to a concentration of 1 unit per 5 µl with water.

Unit Definition

One unit will produce 1.0 μmole of acid soluble polynucleotides from native DNA per min at pH 8.8 at 37 °C, based on EM/260 = 10,000 for the mixed nucleotides.
