Biochem/Physiol Actions

Catalase activity is constant over the pH range of 4.0-8.5. Each subunit of the tetramer contains iron bound to a protoheme IX group. The enzyme also strongly binds NADP, which is in close proximity to the heme group. The enzyme activity is inhibited by 3-amino-1-H-1,2,4 triazole, cyanide, azide, hydroxylamine, cyanogen bromide, 2-mercaptoethanol, dithiothreitol, dianisidine, and nitrate. Incubation of catalase with ascorbate or ascorbate/Cu2+ results in degradation of the catalase molecule.

Application

Catalase agarose has been used in a study to advance the treatment of oxidative stress in neurodegenerative diseases. Catalase agarose has also been used in a study to investigate the growth of cyanobacterium on agar media in the presence of heterotrophic bacteria. Catalases have a wide range of industrial applications. The enzyme is used for the removal of traces of (H2O2) that is added to milk and cheese during sterilization. It is used for the degradation of peroxide used in textile bleaching to prevent any problems in subsequent dyeing. Coupled with oxidases, the enzyme is used to prevent the inactivation of oxidases by the action of high concentration of peroxide. It is also used to prevent side reactions caused by (H2O2) that can degrade the target product.

Caution

Freezing of solutions is not recommended.

One unit will decompose 1.0 μmole of H2O2per min at pH 7.0 at 25 °C, while the H2O2 concentration falls from 10.3 to 9.2 mM, measured by the rate of decrease of A240.
